Псориаз (ПсО), псориатический артрит (ПсА) и метаболический синдром (МС) объединены общими иммуновоспалительными механизмами. МикроРНК рассматриваются как потенциальное молекулярное звено, связывающее воспалительные и метаболические нарушения.</p></sec><sec><title>Цель</title><p>Цель. Оценить экспрессию hsa-miR‑210–3p, hsa-miR‑10b‑5p, hsa-miR‑130a‑3p и hsa-miR‑126–5p у пациентов молодого возраста с ПсО и ПсА с учетом наличия МС.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. В исследование включено 38 пациентов в возрасте до 45 лет с ПсО и 37 с ПсА, из них с МС – 12 и 15 человек, соответственно. Экспрессия микроРНК в плазме определялась методом полимеразной цепной реакции в режиме реального времени (ПЦР–РВ) с последующей обратной транскрипцией, дальнейший анализ проводился по методу Ливака. Анализ экспрессии микроРНК проводился с использованием метода delta-delta ct (2^-ΔΔCt). Для статистической обработки результатов использовали методы PERMANOVA, SIMPER, Т-тест, критерий Манна-Уитни, критерий Шапиро-Уилка, ROC-анализ, корреляционный анализ.</p></sec><sec><title>Результаты</title><p>Результаты. Результаты парного многомерного анализа PERMANOVA для групп ПсО и ПсА продемонстрировали статистически значимые различия совокупного паттерна экспрессии исследованных микроРНК (p=0.003). Статистически значимые различия в экспрессии между пациентами с ПсО и ПсА выявлены для двух микроРНК: miR‑130a‑3p и miR‑10b‑5p: 57,3 % (p=&lt;0.001) и 22,5 % (p=0.021). Наличие МС у пациентов с ПсА сопровождалось повышением экспрессии miR‑10b‑5p (FC=0.68, p=0.03; log2(2^(–dct) ПсА без МС –0.56 [–1.387; –0.198] в сравнении с ПсА с МС –1.465 [–1.842; –0.67], р=0,032). Уровень экспрессии miR‑130a‑3p положительно коррелировал с гликемией, инсулинемией и индексом HOMA-IR.</p></sec><sec><title>Заключение</title><p>Заключение. miR‑130a‑3p и miR‑10b‑5p могут отражать переход от кожной к суставной форме псориатической болезни и участвовать в интеграции метаболических и воспалительных процессов, представляя потенциальные биомаркеры трансформации ПсО в Пс А.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Background</title><p>Background. Psoriasis (PsO), psoriatic arthritis (PsA), and metabolic syndrome (MetS) share common immune-inflammatory mechanisms. MicroRNAs are considered potential molecular links integrating inflammatory and metabolic disturbances.</p></sec><sec><title>Objective</title><p>Objective. To evaluate the expression of hsa-miR‑210–3p, hsa-miR‑10b‑5p, hsa-miR‑130a‑3p, and hsa-miR‑126–5p in young patients with PsO and PsA, taking into account the presence of MetS.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. The study included 38 patients with PsO and 37 with PsA under 45 years of age. Plasma microRNA expression was assessed using RT-qPCR (2^-ΔΔCt method). PERMANOVA, SIMPER, ROC analysis, and correlation analysis were performed.</p></sec><sec><title>Results</title><p>Results. The results of paired multivariate PERMANOVA analysis for the PsO and PsA groups demonstrated statistically significant differences in the overall expression pattern of the studied microRNAs (p=0.003). Statistically significant differences in expression between patients with PsO and PsA were found for two microRNAs: miR‑130a‑3p and miR‑10b‑5p: 57.3 % (p=&lt;0.001) and 22.5 % (p=0.021). The presence of MS in patients with PsA was accompanied by an increase in the expression of miR‑10b‑5p (FC=0.68, p=0.03; log2(2^(–dct) PsA without MS –0.56 [–1.387; –0.198] compared to PsA with MS –1.465 [–1.842; –0.67], p=0.032).
